ABSTRACT

Extracellular vesicles (EVs) represent a promising liquid biopsy platform in multiple myeloma (MM). We developed an MM EV Surface Protein Assay to quantify and dynamically monitor four MM EV subpopulations defined by targetable MM surface proteins (BCMA, CD38, GPRC5D, and CD319) across 336 serial blood samples from 45 relapsed/refractory MM (RRMM) patients treated with anti-BCMA chimeric antigen receptor (CAR) T-cell therapy. All four MM EV subpopulations significantly decreased in 43 patients with initial response, while BCMA+, GPRC5D+, and CD319+ MM EVs increased in 19 patients with progression, and antigen escape was detected by BCMA+ MM EVs. MM EV subpopulations differentiated minimal residual disease (MRD) status and complemented MRD for detecting early relapse before clinical progression. Notably, CD319+ MM EVs were early predictors of progression-free and overall survival in MRD-negative patients. This assay enables noninvasive monitoring of deep response, progression, and antigen escape, and stratifies survival in MRD-negative patients with RRMM.

HarmonyCell: Automating Single-Cell Perturbation Modeling under Semantic and Distribution Shifts

arXiv:2603.01396v2 Announce Type: replace Abstract: Single-cell perturbation studies face dual heterogeneity bottlenecks: (i) semantic heterogeneity--identical biological concepts encoded under incompatible metadata schemas across datasets; and (ii) statistical heterogeneity--distribution shifts from biological variation demanding dataset-specific inductive biases. We propose HarmonyCell, an end-to-end agent framework resolving each challenge through a dedicated mechanism: an LLM-driven Semantic Unifier autonomously maps disparate metadata into a canonical interface without manual intervention; and an adaptive Monte Carlo Tree Search engine operates over a hierarchical action space to synthesize architectures with optimal statistical inductive biases for distribution shifts. Evaluated across diverse perturbation tasks under both semantic and distribution shifts, HarmonyCell achieves a 95% valid execution rate on heterogeneous input datasets (versus 0% for general agents) while matching or even exceeding expert-designed baselines in rigorous out-of-distribution evaluations. This dual-track orchestration enables scalable automatic virtual cell modeling without dataset-specific engineering.

EasyInsert: A Data-Efficient and Generalizable Insertion Policy

arXiv:2505.16187v2 Announce Type: replace-cross Abstract: Robotic insertion is a highly challenging task that requires exceptional precision in cluttered environments. Existing methods often have poor generalization capabilities. They typically function in restricted and structured environments, and frequently fail when the plug and socket are far apart, when the scene is densely cluttered, or when handling novel objects. They also rely on strong assumptions such as access to CAD models or a digital twin in simulation. To address these limitations, we propose EasyInsert. Inspired by human intuition, it formulates insertion as a delta-pose regression problem, which unlocks an efficient, highly scalable data collection pipeline with minimal human labor to train an end-to-end visual policy. During execution, the visual policy predicts the relative pose between plug and socket to drive a multi-phase, coarse-to-fine insertion process. EasyInsert demonstrates strong zero-shot generalization capability for unseen objects in cluttered environments, robustly handling cases with significant initial pose deviations. In real-world experiments, by leveraging just 1 hour of human teleoperation data to bootstrap a large-scale automated data collection process, EasyInsert achieves an over 90% success rate in zero-shot insertion for 13 out of 15 unseen novel objects, including challenging objects like Type-C cables, HDMI cables, and Ethernet cables. Furthermore, requiring only a single manual reset, EasyInsert allows for fast adaptation to novel test objects through automated data collection and fine-tuning, achieving an over 90% success rate across all 15 objects.

Classroom Final Exam: An Instructor-Tested Reasoning Benchmark

CFE-Bench is curated from repeatedly used, authentic university homework and exam problems, paired with reference solutions provided by course instructors. CFE-Bench remains challenging for frontier models: the newly released Gemini-3.1-pro-preview achieves 59.69% overall accuracy, while the second-best model, Gemini-3-flash-preview, reaches 55.46%, leaving substantial room for improvement. Beyond aggregate scores, we conduct a diagnostic analysis by decomposing instructor reference solutions into structured reasoning flows. We find that while frontier models often answer intermediate sub-questions correctly, they struggle to reliably derive and maintain correct intermediate states throughout multi-step solutions. We further observe that model-generated solutions typically contain more reasoning steps than instructor solutions, indicating lower step efficiency and a higher risk of error accumulation.
